Moderna is getting the biggest index promotion of the year. Nasdaq confirmed on Oct 1 that the vaccine maker will enter the Nasdaq-100 before the opening bell on Friday, Oct 9. It takes the slot Warner Bros. Discovery vacates. That exit follows the expected Oct 6 close of the $81 billion Paramount Skydance merger. The momentum behind the stock is extraordinary. Moderna rose 174.98% in the third quarter, finishing September at $192.57 after starting the quarter at $70.03. On the year, the stock is up 544% after closing Oct 2 at $190.01. More than $800 billion in index-tracking products follow the Nasdaq-100, and all of them will need to own Moderna by next Friday.

The rally began with science, not index math. On Aug 19, Moderna and Merck announced that their personalized cancer vaccine had cleared a Phase 3 trial. No mRNA-based cancer therapy had ever done that before.

The trial, INTerpath-001, paired intismeran autogene with Merck's Keytruda. Intismeran autogene — also known as V940 or mRNA-4157 — is a personalized mRNA shot. It was tested in 1,137 patients whose stage IIB to IV melanoma had been surgically removed. It hit its primary goal, recurrence-free survival, and a key secondary goal, distant metastasis-free survival, beating Keytruda alone on both. The companies called it the first Phase 3 success for an individualized neoantigen therapy and the first to show a meaningful edge over Keytruda alone after surgery. It extends five-year follow-up data from the earlier Phase 2b KEYNOTE-942 study, presented at ASCO in June. That study showed a 49% lower risk of recurrence or death and a 59% lower risk of the cancer spreading to distant organs. The market's verdict was instant. On Aug 19, Moderna jumped 177% in one session, from $62.96 to $174.38. Volume hit 199 million shares — roughly twenty times a normal day. "Today's results represent a landmark moment for adjuvant melanoma treatment," said principal investigator Georgina Long of the Melanoma Institute Australia.

Why it matters

Index inclusion turns a stock story into a plumbing story. The Nasdaq-100 is mirrored by more than 200 investment products managing over $800 billion in assets. The managers of those funds do not get to pick and choose. They must hold whatever the index holds, in index weights, before the change takes effect. That mechanical bid lands this week. WBD's removal lines up with the Oct 6 merger close, and Twilio fills its S&P 500 seat the same day. Moderna's Nasdaq-100 debut follows three days later. The familiar pattern for additions is strength into the effective date as passive money accumulates, then choppier trading once the forced buying is finished. For context on the broader rotation, see our indices coverage. Biotech momentum is wider than one name — Iovance posted a 256% third-quarter run of its own.

What to watch

  • Oct 6: Paramount Skydance's $81 billion WBD merger is expected to close; WBD leaves the Nasdaq-100 and S&P 500, and Twilio takes its S&P 500 seat.
  • Oct 9, pre-open: Moderna officially joins the Nasdaq-100; passive rebalancing flows peak around the effective close.
  • Post-inclusion drift: with the stock roughly 8.5 times its 52-week low of $22.28, watch whether index demand fades after Oct 9 — additions often reverse once mechanical buying ends.
  • Pipeline calendar: INTerpath-001 continues toward overall-survival data, and regulators are reviewing the Phase 3 results — the next clinical headline, not the index, decides the longer trend.
  • Analyst positioning: Citi's Geoff Meacham cut MRNA to Sell from Neutral in September (target $80), arguing the roughly 222% run since mid-August had outrun what the pipeline justifies; consensus remains at Hold.

MarketRadar take

Into Oct 9, this tape is driven by flows rather than fundamentals. Index funds must accumulate a stock that is already up 544% on the year. That mechanical bid typically peaks around the effective close. The science underneath is genuine — the first Phase 3 victory for a personalized mRNA cancer vaccine. But the price now discounts a great deal of it. Citi's sell call flags how stretched the 8.5x run from the 52-week low has become. Expect the firmest technical footing into Friday's rebalance, then news-driven trading once the passive bid is done. Our read: flow-positive for MRNA into the event, cautious on chasing beyond it. Without a fresh clinical catalyst, the index pop is borrowed momentum.
